Drug interactions as a preventable cause of iatrogenic disease

Introduction: This review considers the basic principles of drug-drug interactions, risk factors and most commonmechanisms underlying drug-drug interactions. Specific examples of drug-drug interactions describedas clinically relevant are also presented. Methods: We searched Medline, Pubmed and B-on for research articles, reviews and meta-analysis. Referenceworks and the European Medicines Evaluation Agency (EMEA) Note for Guidance were also considered. Overall 97 articles were included. Conclusion: Adverse drug-drug interactions are a major cause of morbidity and mortality. Not all drug--drug interactions can be predicted. However, an increased awareness of the potential for these interactionsto occur and the possibility that signs and symptoms can result from a drug-drug interaction, may allowhealthcare providers to minimize the risk, improve medical assistance and overall contribute to patientsafety.Downloads
